Unicorn, according to the lore, is a mythological creature that has been described since antiquity as a beast with a large, pointed, spiraling horn projecting from its forehead.

Season 2

Dean: (on angels) Because there's no such thing, Sam.

Sam: Dean, there's ten times as much lore about angels as there is about anything else we've ever hunted.

Dean: Yeah, you know what? There's a ton of lore on unicorns too. In fact, I hear that they, they ride on silver moonbeams, and they shoot rainbows out of their ass.

Sam: Wait, there's no such thing as unicorns?

Season 7

An employee of the chain, Howard, is using magic to kill children's parents, with a manifestation of their children's fear. One of the victims of circumstances is a man who is killed when he is impaled by the horn of a unicorn.

Season 9

A unicorn skull could be seen in Magnus' hideout, proving they either do or did actually exist after all.

Trivia

• Unicorns have not yet been presented in the series as a true creature. The unicorn who appears in Plucky Pennywhistle's Magical Menagerie was a manifestation of a child's fear, not a creature.
• Dean does not believe in their existence, but afther see that skul in Magnus home his beliefs may be changed.
• Meg describes Amelia Richardson as a "unicorn" because she made Sam want to quit the hunting life.
