Lenore was a vampire that Sam and Dean encountered in Season 2. Because she didn't drink human blood, she was considered one of the more friendly vampires.

Season 2

Lenore was part of a group of vampires who have sworn off consuming human blood, who instead fed on the blood of cows. She explained to Sam that while she found cow blood disgusting, it is worth it to be able to live in peace and not draw attention to hunters. Despite her efforts, though, she and her nest were being hunted by an obsessed vampire hunter called Gordon Walker, who Sam and Dean had just met while investigating a case. Her group kidnapped Sam from his motel room and brought him to their nest. There, Lenore asks Sam to help the vampires escape and stop Walker from killing them. When Sam is released, he returns to his motel room and pleads the vampires' case to his brother. Gordon overhears him and goes after Lenore's nest on his own. Once the Winchesters realize Gordon overheard them and left to kill them himself, they head towards the nest. There, Gordan has Lenore tied up and has been torturing her with a knife dipped in dead man's blood. When the boys arrive, they attempt to talk Gordon out of killing Lenore, but Gordon argues that all vampires are bloodthirsty killers, even Lenore and her nest. To prove his point, Gordan grabs Sam's arm and cuts it with his knife, and holds his arm above Lenore's face. When his blood drips onto her face, her vampire instincts kick in and she moves towards the blood. But, after a moment, she is able to overcome it, and reverts to normal. With their point proven, Sam escorts Lenore out and helps her leave town. 

Season 6

Years later, Lenore appears in the Season 6 episode "Mommy Dearest" when Castiel brings her from her hiding spot. She is confused at first but notices the boys. Sam asks her for Eve's location, but Lenore wants nothing to do with Eve. Sam also asks about her nest and she tells them they could no longer resist Eve's voice and began killing again. Dean asks again for the location of Eve. Lenore tells them that if she does tell them the location, Eve will know they are coming. The gang still insists, so Lenore finally reveals that Eve is in Grants Pass, Oregon. In return, she asks the brothers to kill her, due to her shame in having relapsed and feeding on a human recently. Sam and Dean are reluctant, but Castiel steps in and incinerates her with his powers.

• She's the first recurring vampire.
• She is, to date, the only known female pack leader; all the other vampire leaders who have been introduced have been men.
• Her actress, Amber Benson, is best known for portraying the witch Tara Maclay in Buffy the Vampire Slayer, which also starred fellow Supernatural guest starts Mercedes McNab , James Marsters , Charisma Carpenter and Harry Groener
• Another possible reason that Cas killed her, was to add an extra soul to purgatory for him to use.
